Abstract
Amaç: Kadınlarda en sık görülen malignite meme kanseridir. Doğal biyoaktif bileşenler, son yıllarda güçlü antikarsinojenik aktiviteleriyle ilgi odağı haline gelmiştir. Klinik çalışmalar, tarçının meme kanserinin önlenmesi ve tedavisi üzerinde olumlu bir etkiye sahip olduğunu göstermiştir. Öte yandan, bitkiler için önemli bir mikro besin maddesi olan borik asidin son zamanlarda bazı kanserleri önleyebileceği bildirilmiştir. Bu çalışmada amaçlanan, tarçın ve borik asidin meme kanseri hücrelerinde apoptozis yoluyla hücrelerin ölümüne neden olup olmadığını araştırmaktır. Bu amaç doğrultusunda, tarçın ve borik asidin tek başına ve birlikte kullanımının hücre büyümesi, hücre ölümü ve apoptotik yola katkıları üzerindeki etkileri incelenmiştir. Bu çalışma, bu doğal bileşenlerin meme kanseri tedavisindeki potansiyellerinin daha iyi anlaşılmasına katkıda bulunmayı amaçlamaktadır. Gereç ve Yöntem: Bu çalışmada, MCF-7 hücre hattı üzerinde; canlılık oranları MTT testi ile tespit edilmiştir, IC50 dozları belirlenmiştir ve Bax, Kalpain, Sitokrom C ve p53 düzeyleri ELISA yöntemi ile analiz edilmiştir. Bulgular: Tarçın ve borik asit kombinasyonunun meme kanseri hücrelerinde apoptozu güçlü bir şekilde etkilediği tüm markerlarda gözlemlenmiştir. Sonuç olarak, salt tarçının olumlu etkileri yalnızca p53'te gözlemlenmiştir. Salt borik asidin, Sitokrom C'de ve p53'te olumlu etkileri gözlemlenmemiştir. Tarçın ve borik asit kombinasyonunun olumlu etkileri ise yalnızca Sitokrom C'de gözlemlenmiştir. Sonuç: Tarçının ve borik asidin MCF-7 hücre hattında hücre canlılığını azalttığı gözlemlenmiştir ve tarçın ve/veya borik asidin bazı markerlarda apoptoza olumlu etkilerine dair sonuçlar elde edilmiştir. Çalışmadan elde edilen bu sonuçlar, başka modellerde de incelenebilecek ve edinilen sonuçların kliniğe uygulanabilmesi için yapılacak çalışmalara ışık tutacaktır. Bu sonuçlar, bu doğal bileşiklerin meme kanseri tedavisinde potansiyel bir alternatif strateji olarak değerlendirilmesini önermektedir. Objective: Breast cancer is the most common malignancy in women. Natural bioactive compounds have recently gained attention due to their potent anticarcinogenic activities. Clinical studies have shown that cinnamon positively affects the prevention and treatment of breast cancer. On the other hand, boron, an essential micronutrient for plants, has been reported to have potential preventive effects against certain types of cancer. This study investigated whether cinnamon and boric acid induce cell death through apoptosis in breast cancer cells. The study examined the effects of cinnamon and boric acid, alone and in combination, on cell growth, cell death, and their contributions to the apoptotic pathway. This research aims to contribute to a better understanding of the potential of these natural compounds in breast cancer treatment. Materials and Methods: In this study, the ELISA method was used to look at the levels of Bax, Calpain, Cytochrome C, and p53 in the MCF-7 cell line. The MTT assay was used to find out how alive the cells were. Results: All observed markers showed that combining cinnamon and boric acid strongly influenced apoptosis in breast cancer cells. The positive effects of cinnamon were only observed in p53. However, the positive effects of boric acid were not observed in cytochrome C and p53. The positive impact of the combination of cinnamon and boric acid was only observed in cytochrome C. Conclusion: Cinnamon and boric acid decreased cell viability in the MCF-7 cell line, and positive effects on apoptosis were observed in specific markers with cinnamon and boric acid. These results can be further investigated in other models and provide insights for future studies aiming to apply the obtained results in clinical settings. These findings suggest that these natural compounds should be considered potential alternative strategies for treating breast cancer.
